Trimethoprim and Sulphadiazine had a synergistic effect. Trimethoprim is a dihydrofolate reductase inhibitor. It inhibits the conversion of bacterial dihydrofolic acid to tetrahydrofolic acid, which is necessary for the synthesis of certain amino acids, purines, thymidine, and ultimately DNA synthesis. Trimethoprim may be bacteriostatic or bactericidal against a wide range of gram negative and gram-positive aerobes, as well as some protozoa. Sulphadiazine Sodium have a similar structure to P-amino benzoic acid and to interfere with the synthesis of nucleic acids in sensitive microorganism by blocking the conversion of P-aminobenzoic acid to the co-enzyme dihydrofoloic acid, a reduced form of folic acid. Their action is primarily bacteriostatic, although they may be bactericidal where concentrations of thymine are low in the surrounding medium. The sulphonamides have a broad spectrum of action.
